封闭及免疫杂交 为了防止抗体和膜非特异性的结合,要将除目的蛋白外其他无结合蛋白位置封闭起来,防止后面一抗非特异性结合产生较深背景。常用封闭物有脱脂奶粉和 BSA(稀释于不同的缓冲液中),但不同的抗原-抗体反应,封闭条件均不相同。 封闭后,目标蛋白便与特异性的一抗进行免疫反应,一抗可以是标记的,也可以是非标记的。标记的一抗与免疫原结合后,可直接进行检测(直接法),降低了背景和非特异性结合,但信号较弱。非标记的一抗配合标记的二抗使用(间接法),对信号有级联放大作用,可以增加灵敏性和信噪比。二抗既可标记放射性同位素,也可标记染料或其他分子,或与酶偶联。常用的酶包括碱性磷酸酶(AP)和辣根过氧化物酶(HRP),通过与底物反应产生光信号。 用适量的TBS/PBS洗已经转好的膜,室温,放到摇床上晃动。 1. 将膜置于约25 ml封闭缓冲液中1 h,室温,置于摇床上晃动。 2. 15 ml TBS/T或PBS/T洗膜三次(每次约5 min)。 3. 加入稀释过的一抗,室温孵育1-2 h或4 ℃过夜,缓慢晃动。 4. 用15 ml TBS/T或PBS/T洗膜三次(每次约5 min)。 5. 加入稀释过的二抗,室温孵育1 h,缓慢晃动 6. 用15 ml TBS/T或PBS/T洗膜三次(每次约 5min)。(注:最后一次要用不含Tween的溶液清洗) 注:实验的条件可根据实验情况进行调整。
